The Relationship between the Effects of Nitric Oxide and BK Channels on Proliferation of Human Dermal Fibroblasts

|
Nitric oxide (NO) regulates fibroblast proliferation and apoptosis and skin homeostatic functions. Large conductance Ca2+-activated K+ (BK) channels have been associated with the regulation of NO production and cell proliferation. This study was undertaken to investigate the effects of NO and BK channels and their relationship in the proliferation of human dermal fibroblasts by electrophysiological patch-clamp techniques and cell proliferation assay. NS1619 (a specific BK channel activator) increased the activity of the K+ channels in whole-cell and single-channel mode patch clamp recordings. However, iberiotoxin (IBTX, a specific BK channels blocker) decreased the activity of the channels. S-nitroso-N-acetylpenicillamine (SNAP, a specific NO donor) activated the IBTX- sensitive K+ channels in whole-cell and single-channel recordings, and showed biphasic effects on cell proliferation which were dependent on the concentration and duration of culture in cell proliferation assay. To evaluate how BK channel modulators affect on NO stimulation effects for cell proliferation, we added them on the cell culture with SNAP (100¥ìM) until 24 hr. However, SNAP-induced increase of fibroblast proliferation for 6¡24 hr in 48 wells was not affected by BK channel modulators, NS1619 (50¥ìM) or IBTX (100 nM). These results suggest that BK channel may not be involved in the SNAP-induced stimulation effect on the proliferation of human dermal fibroblasts.
